Biotinylated Long R3 IGF- I is prepared by a selective biotinylation procedure of an 83 amino acid analog of human IGF-I comprising the complete human IGF-I sequence with the substitution of an Arg (R) for the Glu (E) at position 3 (hence R3), and a 13 amino acid extension peptide at the N-terminus (Francis, et al., J. Mol. Endocrinol. 8: 213, 1992).. As in the non-biotinylated IGF-I analog it is the IGF-I analog with the lowest affinity to IGFBP´s, but it binds to receptors. No binding to IGFBP-6 was detected in our western-ligand blot.
The Long R3 IGF-I is derived from bacteria (E.coli) and biotinylation was performed by chemical synthesis.
Formulation: Lyophilised from a solution of 0.5 ml TBS, pH 7.4, containing 10 µg of the biotinylated product . A preservative (0.05 % ProClin 150) and a stabilising protein of non-mammalian origin are added to ensure stability of the reconstituted product.
Recommended dilution: 1: 100 for western ligand blotting. Tested with our western ligand blotting kit reagents.
Storage: Store refrigerated upon arrival. The product is stable at 4 °C for at least one year. Reconstitute with 0.5 ml water and store aliquots frozen at - 20 °C. Avoid repeated freeze-thawing cycles.
