Anti-human IGF-II Polyclonal Antiserum Specifications

The Product: Anti-human IGF-II is a polyclonal antiserum specific
for human, rat, chicken and porcine IGF-II amongst the species tested.
Cross reactivity with either ovine, insulin, human, rat or chicken
IGF-I is undetectable. This polyclonal antiserum is suitable for use
in radioimmunoassay, immunoblotting and immunoprecipitation.
Source: The antiserum is derived from rabbit serum raised
against recombinant human IGF-II ovalbumin conjugate.
Quantity: 20µl of serum
Appearance: White Powder
Specificity: The antiserum is specific for human IGF-II.
Cross-reactivity with porcine, chicken and bovine IGF-II is 100 %
and 85% respectively. Less than 0.1 % cross-reactivity is
observed with ovine, insulin, human, rat and chicken IGF-I.
Titre: At a dilution of 1/7500 the antiserum binds 25% of
[125I]-labelled human IGF-I tracer (80Ci/g). Half-maximal inhibition
(ED50) of radioligand binding is produced by 600pg of human
IGF-II.
Reconstitution: Dissolve in a suitable buffer solution appropriate
for desired application.
Storage/Stability: At least 2 years at 2-4°C (lyophilised). After
reconstitution store at -20°C.
Radioimmunoassay
For radioimmunoassay (RIA), the recommended dilution of
antiserum binds 25% of fresh [125I]-labelled human IGF-II tracer
(80 Ci/g). Half-maximal inhibition (ED50) of radioligand binding is
produced by 600 pg of human IGF-I and the limit of detection is
approximately 20-30 pg.
The relative potencies of the following peptides at inhibiting the
binding of [125I]-labelled human IGF-II to this antiserum were found
to be:
human IGF-II: 100%
human IGF-I: <0.1%
chicken IGF-II: 100%
chicken IGF-I: <0.1%
rat IGF-II: 100 %
rat IGF-I: <0.1%
porcine IGF-II: 100%
bovine IGF-II: 85%
bovine insulin: <0.1%
